Bryant scored 27 points and O'Neal, playing on his home court, tallied 17 points on 8-of-9 shooting, helping the Western Conference to a 146-119 thrashing of the East at US Airways Center.
The dynamic duo played together for the Los Angeles Lakers from 1996-2004, winning three championships until O'Neal was dealt to Miami. Last February, three days after the All-Star contest, O'Neal was traded back to the Western Conference as he joined the Phoenix Suns.
The two were named as co-MVPs of the All-Star Game, with each garnering a third MVP honor for the contest.
"I'm just happy to be here, it's probably my last one," said O'Neal. "At 36, I'm just glad the coaches voted me in. I'm just happy Kobe was looking for me today, other guys were looking for me today."
Bryant was the MVP of the game in 2002 and '07, while O'Neal won it in 2004 and was co-MVP with San Antonio's Tim Duncan in 2000.
It's the first time Kobe and Shaq have played together since 2004, and they were coached by current Lakers coach Phil Jackson.
"It was fun," said Bryant. "Phil diagramed a few things for us to play a two- man game like we used to. We went back to what we were used to doing, so it was fun."
New Orleans guard Chris Paul had 14 points, 14 assists and seven rebounds for the West, while Phoenix's Amare Stoudemire scored 19.
Cleveland's LeBron James had 20 points for the East, which also received 18 apiece from Miami's Dwyane Wade and Boston's Paul Pierce.

Chicago Bears (+350) - The Bears are probably a bit of an overhyped team once again this year.No, we don't think it is plausible for QB Jay Cutler to have as bad of a season as he did last year, and we do think the additions of RB Chester Taylor and DE Julius Peppers are going to help immensely, but there's still something in the water in the Windy City that we aren't so sure about. Maybe Chicago finds its way to .500... but then again, maybe it doesn't. If Favre comes back, Cutler might be the worst quarterback in this division this year.
Detroit Lions (+1500) - The Lions are probably once again going to be the whipping boys for the rest of the teams in the NFC North, but they aren't just going to roll over and die once again. There is some real talent amassing on this team offensively, as the combination of QB Matt Stafford, RB Jahvid Best, WR Calvin Johnson, and TE Brandon Pettigrew should put a bunch of points on the board if the offensive line can even remotely hold up. The questions really lie on defense, where it feels like DT Ndamukong Suh is going to be trying to stop opposing teams all by himself. This year should show some marked improvement in the Motor City.
